With nothing around you for roughly 100 ft..doors closed,not power poles n trees ect..Radio on ch 1..set switch to cal..Key mic and turn calibration knob to the right till needle cal..Release mic,go back to cal switch and change to swr..Key up again and make note of what swr setting is at...release mic...Do that in ch 1 and 40...That will tell you if ant need adjustment or not....Try not to key up to many times and do it quickly....

Hey man this actually just happened to me a few days ago. I had a cobra 29 my uncle gave to me, and I could hear drivers fine they just couldn’t hear me. Anytime I keyed the mic they got rf but nothing came through. The calibration knob itself took a #### on me and I couldn’t even turn the dial in either directions to calibrate. Borrowed a friends mic thinking mine was broken but still nothing; so I bought a cobra 29 Ltd chrome and everything works awesome!

You need an external swr/power meter to see if the radio is putting any RF out. Otherwise it's just a guessing game.
Have you tried a different microphone like was mentioned to do? That would be the first and easiest thing to do.Life on Wheels Thanks this. -
Those cheap meters are not much better

I have a 29LTD, and the key on the stock mic sometimes sticks when pressed. It feels right, but no TX. New mic is on my shopping list.
