They will check your eyesight(with your corrective lenses on), your blood pressure, your pulse rate, your relexes and peripheral vision and listen to your heart and lungs and also check your hearing. They will also check to see if you are color blind. They will take a urine sample too but not for drugs. I think they ckeck it for substances that would indicate that there may be a problem with your blood sugar, at which point if they did find something I'm pretty sure that they would draw some blood. If you are okay in all of these areas, you'll be good for 2yrs.

Go to DOT's website and search for "medical examination report". You will find the first result will be a pdf file with everything the doctor is supposed to check.
