Actually it depends on the station.
You've gotten a lot of correct answers. But there are exceptions to your question, which was vague at best.
Vehicle Inspection Station/Area come in many forms. Some are located within a State Scale and you only enter the "inspection" area when asked to, some are simply wide spots that officers flag the truck down they want to inspect. Others direct ALL trucks to enter, and they grab one from the line.
Best advice, pay attention to all signs. And wake your sorry ### trainer up, and ask him. That's what he's supposed to be teaching you.
Regardless of what it's called,
Port Of Entry,
Weigh Station,
Inspection Station,
Agricultural Check point,
Border Patrol Check Point,
Customs.
You need to plan on stopping, unless directed to do otherwise.

PA used to have them on the turnpike...don't know anymore don't go pass the Indiana/Ohio line...that's East Coast paid my dues and I'm not going back.
IL will have them set up in Rest Areas (temporary they move day to day). AL has vehicle inspection areas....don't worry they post a sign just go in and smile don't worry they'll do the rest. MO has them but they seem to move on a daily basis...must be why we call that state Misery.
Also never ever blow past a VA scale they don't care if you're lined up a mile away get on the shoulder and wait. They will come after you and they are always open.....okay they use to be open....another East Coast state I don't go to.
If the sign says vehicle inspection it means you not a 4-wheeler and yes they will chase you down and give you a safe driving award that is going to hurt the pocket. -
NY does them in rest areas but most places have a big sign with blinky lights when open. I ran into an agricultural check in FL last summer but they laughed at me and waved me through, maybe because I had a load of iron pipe? What's the point of the ag check anyway?

Many years back, AZ & CA tried to prevent infestation of their states by fire ants.
If you showed up at a check point and they found even 1 dead ant. They would fumigate the load, or turn you around.
We see how well that worked -
WhenI was delivering RV's with a 1ton I asked at a FL ag check if I was required to stop. They said I was because I was commercial . Private RV's didn't have to .That made no sense .

They use rest areas sometimes as well in some states, although i can't speak for MO i've never seen one there i was stopped in North Dakota a few times, and in Ohio once.

Sometimes Colorado sets up temporary inspection sites on get-off ramps. There will be a temporary sign set up a few miles in advance. But they have a trooper standing there motioning you in or to pass on by. I don't recall seeing this in other states, but I am sure they exist. I'm with Dance, though... WAKE UP YOUR TRAINER! You shouldn't have to ask this question on this forum unless he doesn't know the answer either... which wouldn't surprise me with some of the "trainers" we have out here on the road.
