Can anyone answer what might cause this code 72. The code is 1 and 2 cylindr fault code. It only happens for a few seconds then the truck runs fine for 500 miles then acts up again for a few sec.

Code 72 are random misfire in cylinders 1&2. Possibly caused my many different things.
Ran fuel cleaner, still misfiring,Check glow plugs, Exhaust, Header, underdrive pulleys, grounding wires.

You have a common wire that is broken between the ECM and injectors 1 & 2. When you lose current at this wire cylinders 1 & 2 will misfire until continuity is restored. It could be the plug at the wiring harness connection at the valve cover, start wiggling wires with the engine running until you make it misfire..........then you know you're close to it. You should be able to isolate it to which wire it is and where with wiggling each wire independently..........it should be a white wire

Thanks guys for your advice, its very well appreciated. Now I will know what to look for...one other thing. When it does this it loses power like if I have a clogged fuel filter. Could this possibly also cause this to happen?

I had the same problem when I brought the rpms above 1700 on a pull. I had to replace the hardness INSIDE the valve covers. Can't remember if it cost around 350.00, for part only. And had to call several dealers to find it.

I've replaced that harness...its two of them. They run around 165.00 a piece, I replaced both of them. I was getting a code 95 and they do a repair kit on the harness. I decided to replace it. This was a few agos.

That is an old design turbo with the S400 frame. It has a small shaft and bearings and isn't woth the time to install. I prefer the newer S410 frame turbos with the Extended Tip Technology compressor wheels.
