Is it possible to start off with a least through Ryder or Penske if you don’t have start up cash?

I wouldn't recommend it. I would advise saving some additional money on the front end as it makes things much easier down the road.

Cash for the truck/trailer is only half the battle. Insurance and other fees will cost as much or more than the cost of the rig.

Both Ryder and Penske have frieghtliners those are good for getting your feet wet... ecm fuse box and rug gets wet too

You can do the lease thing with a trucking company but they are rip off and almost set you up to fail. The No Money down walk away lease. Everyone does them for the same reason it seems. They don't have any money but want to drive their own truck.
You can get a truck from Loan Mountain with some money down and it is your truck to keep. That means when it breakdown you got pay to fix it and you can't just walk away. -

You have to be in business for 3 years minimum before Ryder will even consider you for a lease.

My guess, based only on the question you posted, is that you do not have a business plan. Outside of a company lease, you’re going to have a hard time finding lease programs with out one. Most are going to want some cash up front as well as seeing some operating cash.
